To explain: The use of Flow Cytometry in the diagnosis of leukemia.

Introduction: Flow Cytometry is an approach which sorts different cell type. The cell mixture is stained with particular dyes coupled with fluorescently labeled antibodies. The mixture of the cells is allowed to pass through a tiny liquid stream through a beam of laser light. The fluctuations in the intensity of light and the color of fluorescence are analyzed. Based on these observations each cell is sorted according to their DNA content, surface proteins, and morphology.

In B-cell lymphoma, B-cells are known to express variations in their morphology as well as the expression of various surface receptors example CD19. CD19 is a significant cell surface receptor. It is expressed by all the B-cell population, at every stage of development.

If a human is expected to have B-cell lymphoma then flow- cytometry can be used as a diagnostic tool by doing the following experiments:

  • Performing immunostaining of the cells with anti-CD19 antibody and one other fluorescent dye which absorb at a different wavelength.
  • By determining the number of cells in the S(synthesis)/M (metaphase) of the cell cycle which are also CD19 positive

If a higher number of CD19 positive cells are present in the S (synthesis phase) and M (metaphase) of the cell cycle. then this observation shows that these cells are proliferating on a high rate and high number of B-cells indicates the condition of B-cell lymphoma.

Hence, the above observations are an indication of B-cell lymphoma condition.
