Chapter 10.1, Problem 1TQ
Summary Introduction

To review:

The following when fljB has been deleted but fljA is still expressed in a cell:

1. The motility of cells.

2. The type of flagella produced.

3. The phase variation of cells.

4. The deletion of gene fliC.

Introduction:

Phase variation is a process in which the organism changes its shape and immunological appearance with the help of gene regulation so that the pathogen cannot be recognized by the antibodies. This leads to the wastage of antibodies produced for the original configuration of the pathogen and affects the organism. It is of two types: gene inversion and slippage-strand mispairing.

Explain the concept of loss of heterozygosity (LOH). Why do most cancer cells exhibit LOH of one or more genes? How does failure of the spindle assembly checkpoint lead to loss of heterozygosity?
The localization of Ase1 and the phenotype of its deletion imply that this protein binds to anti-parallel Microtubules. What would you expect for the organization of a protein that has this property?
When the gene encoding a certain cellular kinase is deleted, the resulting mutant cells arrest in the cell cycle and do not divide. These mutants can be rescued when a gene encoding an N-terminal GFP fusion of the protein is expressed, but not when a gene encoding a C-terminal GFP fusion is expressed. Which fusion protein is appropriate to use in studying cellular localization and activity? Explain why.
